Difacta (Emtricitabine + Dolutegravir + Tenofivir Alafemide)

Difacta is a fixed-dose combination antiretroviral therapy containing Emtricitabine, Dolutegravir, and Tenofovir Alafenamide (TAF), designed for the treatment and management of HIV-1 infection in adults and adolescents. This FDA/DOH/WHO-recommended. This triple-drug regimen combines an integrase strand transfer inhibitor (INSTI) with two nucleoside reverse transcriptase inhibitors (NRTIs) for comprehensive viral suppression.

**How It Works**

The synergistic action of these three agents targets HIV replication at multiple points in the viral life cycle. Dolutegravir inhibits integrase, preventing viral DNA integration into host cells. Emtricitabine and Tenofovir Alafenamide block reverse transcriptase, interrupting viral RNA conversion and reducing overall viral load. Together, they support immune reconstitution and help achieve and maintain undetectable viral loads (below 50 copies/mL).

**Dosage & Administration**

One tablet orally once daily with or without food. The single daily dose improves medication adherence and simplifies treatment regimens for long-term HIV management.

**Common Side Effects**

Nausea, diarrhea, headache, fatigue, dizziness, and sleep disturbances may occur initially. Most side effects are mild, transient, and diminish as the body adjusts.

**Serious Considerations**

Rare but serious effects include hepatotoxicity and renal impairment. Regular laboratory monitoring (liver function tests, renal function, CD4 count, and viral load) is essential for safety and efficacy assessment.
