Linadex (Lenalidomide)

Lenalidomide is an oral immunomodulatory drug (IMiD) derived from thalidomide. It has antineoplastic, anti-inflammatory, and immunomodulatory effects. The drug works by enhancing T-cell and NK-cell mediated immunity, inhibiting angiogenesis, and directly suppressing tumor cell proliferation.

Main Uses

• Multiple Myeloma (MM) – usually combined with dexamethasone or other agents.
• Myelodysplastic Syndromes (MDS) – especially in patients with deletion 5q cytogenetic abnormality.
• Mantle Cell Lymphoma (MCL) – in relapsed or refractory cases.
• Follicular Lymphoma (FL) and Marginal Zone Lymphoma (MZL) – in some treatment protocols.

Contraindications

• Pregnancy – Lenalidomide is highly teratogenic (causes severe birth defects). Strict pregnancy prevention and monitoring programs are required.
• Hypersensitivity to lenalidomide or any component of the formulation.
• Breastfeeding – contraindicated due to potential risk to infants.
• Caution in patients with:
  • Severe renal impairment (dose adjustments needed).
  • History of thromboembolic events (increased risk of DVT/PE, especially when combined with corticosteroids or chemotherapy).
