Paloset (Palonosetron)

Palonosetron is a selective serotonin (5-HT3) receptor antagonist that blocks the action of serotonin, a natural chemical in the body responsible for triggering nausea and vomiting. As a second-generation 5-HT3 antagonist, it offers a longer half-life and stronger receptor binding than older agents like ondansetron, making it particularly effective in preventing both acute and delayed nausea and vomiting.

Main Use

• Prevention of nausea and vomiting associated with:
  • Chemotherapy (both acute and delayed phases, especially in moderately to highly emetogenic chemotherapy)
  • Postoperative nausea and vomiting (PONV)

Contraindications

• Known hypersensitivity to Palonosetron or any component of the formulation.
• Caution is advised in patients with:
  • History of hypersensitivity reactions to other 5-HT3 receptor antagonists.
  • Electrolyte imbalances (since they can increase the risk of QT prolongation).
  • Pre-existing arrhythmias or those taking other QT-prolonging medications
