Voriconazole is a second-generation triazole antifungal medication trusted for treating serious, life-threatening fungal infections resistant to first-line therapies. Available as Vorica, this prescription antifungal provides broad-spectrum coverage against Aspergillus, Candida, and other invasive fungal pathogens.
How it works: Voriconazole inhibits fungal lanosterol 14α-demethylase, disrupting ergosterol synthesis in fungal cell membranes. This mechanism prevents fungal growth and replication, making it highly effective against azole-resistant strains.
Clinical applications include invasive aspergillosis, candidiasis (including oesophageal and disseminated forms), scedosporium infections, and other serious systemic mycoses. Particularly valuable for immunocompromised patients, including those undergoing chemotherapy or solid organ transplantation.
Key benefits: Superior CNS penetration compared to fluconazole, making it effective for fungal meningitis. Long half-life supports convenient twice-daily dosing. Available in multiple formulations—oral tablets, suspension, and intravenous injection—ensuring flexible treatment options.
Important considerations: Visual disturbances (photopsia, blurred vision) occur in 20–30% of patients but typically resolve. Monitor liver function regularly, as hepatotoxicity is possible. Drug interactions are significant; voriconazole inhibits CYP3A4 and CYP2C19. Not recommended in pregnancy without clear medical necessity.
Dosing: Standard induction 6 mg/kg IV twice daily for one day, followed by 4 mg/kg twice daily. Oral maintenance typically 200 mg twice daily in adults.
Store in a cool, dry place away from direct sunlight. Dispense in original container with child-resistant cap. Keep out of reach of children and pets.
Always use under strict medical supervision. Report any visual changes, abdominal pain, jaundice, or unusual bruising immediately to your healthcare provider.
